Never can cryopreserved cells be kept at -20 °C.
EDTA is used to chelate free magnesium and calcium ions in order to maintain trypsin inhibition. It is recommended that cells be washed with EDTA prior to trypsin treatment to eliminate all divalent ions from the medium.
