Description
IL-17 is a potent proinflammatory cytokine produced by activated memory T cells. There are at least six members of the IL-17 family in humans and in mice. As interleukin-17 shares properties with IL-1 and TNF-alpha, it may induce joint inflammation and bone and cartilage destruction. This cytokine is found in synovial fluids of patients with rheumatoid arthritis, and produced by rheumatoid arthritis synovium. It increases IL-6 production, induces collagen degradation and decreases collagen synthesis by synovium and cartilage and proteoglycan synthesis in cartilage. Interleukin-17 is also able to increase bone destruction and reduce its formation. Blocking of interleukin-17 with specific inhibitors provides a protective inhibition of cartilage and bone degradation. Recombinant human IL-17 produced in HEK cells is a glycosylated homodimer, having a molecular weight range of 30-35 kDa due to glycosylation. It has been purified by proprietary chromatographic techniques.

                            Activity
                                rHu IL-17 is fully biologically active when compared to standards. The specific activity was determined by the dose-dependent induction of IL-6 secretion from NDHF adult fibroblasts and is typically 5-25 ng/ml.

                            Formulation
                                Sterile-filtered, white, lyophilized (freeze-dried)powder. Lyophilized from a concentrated protein solution (1 mg/ml) in 1xPBS.
                            Reconstitution
                                Please centrifuge product briefly before opening vial. The lyophilized protein should be reconstituted in sterile, ultra-pure water to a concentration of 0.1 - 1.0 mg/ml. This solution can then be diluted into other aqueous buffers and stored at -20°C for future use.
                            Storage & Stability
                                The lyophilized protein, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted rHu IL-17 should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long-term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid repeated freeze-thaw cycles.
