Description
Recombinant Human Interleukin-12 (rHu IL-12) is produced by activated B-lymphocytes, macrophages and connective tissue type mast cells. It is a potent regulator of cell mediated immune responses and induces IFN-γ production by NK and T cells. Among its biological activities IL-12 promotes the growth and activity of activated NK, CD4+ and CD8+ cells and induces the development of IFN-γ producing Th1 cells. Recombinant human IL-12 produced in CHO cells is a 75.0 kDa heterodimeric glycoprotein consisting of disulfide-linked 35 kDa (p35) and 40 kDa (p40) subunits (503 total amino acid residues).

                            Activity
                                Assay #1: Determined by its ability to induce IFN-γ production from NK cells co-stimulated with IL-18. The expected ED50 is ≤ 1.0 ng/ml, corresponding to a specific activity of Greater than 1×10^6 IU/mg. 
Assay #2: Determined by its ability to increase IFN-γ production by anti-TCR mAb-stimulated PBMCs. The expected ED50 for this effect is 4.0-8.0 ng/ml.

                                Less than 1 EU/μg.
                            Formulation
                                Sterile filtered through a 0.2 micron filter and lyophilized from 0.5x PBS (pH 7.2).
                            Reconstitution
                                Please centrifuge product briefly before opening the vial. The lyophilized IL-12 should be reconstituted in sterile 1x PBS (pH 7.2-7.4) to a concentration of 0.2 mg/ml. This solution can then be diluted into other aqueous buffers and stored at -20°C for future use.
                            Storage & Stability
                                The lyophilized powder, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted rHu IL-12 should be stored long-term in undiluted working aliquots at -20°C. For long-term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid repeated freeze / thaw cycles.
