Description
IL-17 is a potent proinflammatory cytokine produced by activated memory T cells. There are at least six members of the IL-17 family in humans and in mice. As Interleukin-17 shares properties with IL-1 and TNF-alpha, it may induce joint inflammation and bone and cartilage destruction. This cytokine is found in synovial fluids of patients with rheumatoid arthritis, and produced by rheumatoid arthritis synovium. It increases IL-6 production, induces collagen degradation and decreases collagen synthesis by synovium and cartilage and proteoglycan synthesis in cartilage. Interleukin-17 is also able to increase bone destruction and reduce its formation. Blocking of Interleukin-17 with specific inhibitors provides a protective inhibition of cartilage and bone degradation. Recombinant murine IL-17 produced in E. coli is a homodimeric, non-glycosylated polypeptide chain of 266 amino acids and a molecular mass of 29,956 Dalton.

                            Activity
                                Recombinant mouse IL-17 is fully biologically active when compared to a standard. The ED50 as determined by the dose-dependent induction of IL-6 production in cultured mouse NIH 3T3 fibroblast cells was found to be 0.4 ng/ml corresponding to a specific activity of 2.5 x 10^6 units/mg.

                            Formulation
                                Lyophilized (freeze-dried), white powder. Lyophiliz ed from a concentrated (1 mg/ml) protein solution containing no additives.
                            Reconstitution
                                Please centrifuge product briefly before opening vial. The lyophilized protein should be reconstituted in sterile, ultra-pure water to a concentration of 0.1 - 1.0 mg/ml. This solution can then be diluted into other aqueous buffers and stored at -20°C for future use.
                            Storage & Stability
                                The lyophilized protein, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted rMu IL-17 should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long-term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid repeated freeze-thaw cycles.
