Description
BAFF, a member of the TNF family of ligands, is expressed in T cells, macrophages, monocytes and dendritic cells. BAFF is involved in stimulation of B and T cell function, and is an important survival and maturation factor for peripheral B cells. BAFF signals through three different TNF receptors TACI, BCMA and BAFF-R. Using specific mAbs, BAFF-R was found to be the predominant BAFF receptor expressed on peripheral B cells, in both humans and mice, and antagonist mAbs to BAFF-R blocked BAFF-mediated costimulation of anti- micro responses. The other BAFF receptors showed a much more restricted expression pattern, suggestive of specialized roles. Recombinant human B lymphocyte stimulator produced in E. coli is a single, non-glycosylated polypeptide chain of 153 amino acids and a molecular mass of 17,007 Dalton.

                            Activity
Recombinant B Lymphocyte Stimulator is fully biologically active when compared to a standard. The activity is determined by a mouse splenocyte survival assay. The ED50 for this effect is 0.5 - 2.0 μg/ml.
                            Endotoxin Level
                                Less than 1 EU/μg.
                            Formulation
White (freeze-dried) powder. Lyophilized from a sterile-filtered protein solution in PBS (pH 7.0).
                            Reconstitution
Please centrifuge product briefly before opening vial. The lyophilized protein should be reconstituted in sterile, ultra-pure water to a concentration of 0.1-1.0 mg/ml. This solution can then be diluted into other aqueous buffers and stored at -20°C for future use.
                            Storage & Stability
The lyophilized protein, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted rHu BAFF should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long-term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid repeated freeze-thaw cycles.
