Description
The three mammalian isoforms of TGF-β, TGF-β1, β2, β3, signal through the same receptor and elicit similar biological responses. They are multifunctional cytokines that regulate cell proliferation, growth, differentiation and motility as well as synthesis and deposition of the extracellular matrix. They are involved in various physiological processes including embryogenesis, tissue remodeling and wound healing. They are secreted predominantly as latent complexes which are stored at the cell surface and in the extracellular matrix. The release of biologically active TGF-β isoform from a latent complex involves proteolytic processing of the complex and /or induction of conformational changes by proteins such as thrombospondin-1. TGF-β2 has been shown to exert suppressive effects on IL-2 dependent T-cell growth, and may also have an autocrine function in enhancing tumor growth by suppressing immuno-surveillance of tumor development. Recombinant human TGF-β2 produced by transient expression of TGF- β2 in non-transgenic plants contains a 6-His-tag at the N-terminal end and is purified by sequential chromatography (FPLC). The product does not contain any animal-derived components or impurities. Recombinant human TGF-β2 is a 27.08 kDa protein composed of two identical 118 amino acid polypeptide chains linked by a single disulfide bond.

                            Activity
The biological activity of TGF-β2 was measured in culture by its ability to inhibit the mink lung epithelial (Mv1Lu) cells proliferation. The ED50 was found to be ≤50 ng/ml.
                            Endotoxin Level
                                Less than 0.04 EU/μg.
                            Formulation
                                Lyophilized from a 0.05 M Tris-HCl buffer (pH 7.4).
                            Reconstitution
Please centrifuge product before opening the vial. The lyophilized protein should be reconstituted in water to a concentration of 25-50 μg/ml. This solution can then be diluted into other aqueous buffers and stored at -20°C for future use.
                            Storage & Stability
The lyophilized protein, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted protein should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long-term storage, it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid repeated freeze-thaw cycles.
