Recombinant Human Interleukin-10 (rHu IL-10), originally known as Cytokine Synthesis Inhibitory Factor (CSIF), is an 18.6 kDa protein of 161 amino acid residues that shares over 80% sequence homology with the Epstein-Barr Virus protein BCRFI. The reported biological activities of rHu IL-10, which may be interrelated, include inhibition of macrophage-mediated cytokine synthesis, suppression of the delayed-type hypersensitivity response, and stimulation of the TH2 cell response, which results in elevated antibody production. IL-10 is a cytokine produced primarily by monocytes and to a lesser extent by lymphocytes. This cytokine has pleiotropic effects in immunoregulation and inflammation. It down-regulates the expression of Th1 cytokines, MHC class II Ags, and costimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. This cytokine can block NF-kappa B activity, and is involved in the regulation of the JAK-STAT signaling pathway. Knockout studies in mice suggested the function of this cytokine as an essential immunoregulator in the intestinal tract. Recombinant human IL-10 produced in HEK cells is a glycosylated non-disulfide bonded homodimer, having a total molecular weight of 17 kDa. It has been purified by proprietary chromatographic techniques.

                            Activity
                                Human recombinant IL-10 is fully biologically active when compared to a standard. The specific activity was determined by the dose-dependent stimulation of the proliferation of MC/9 cells (mouse mast cell line) in the presence of 200 pg/ml IL-4, and is typically 0.3-1.5 ng/ml.

                            Formulation
                                Sterile-filtered, white, lyophilized (freeze-dried)powder. Lyophilized from a concentrated protein solution (1 mg/ml) in 1xPBS.
                            Reconstitution
                                Please centrifuge product briefly before opening the vial. The lyophilized rHu IL-10 should be reconstituted in sterile, ultra-pure water to a concentration of 0.1-1.0 mg/ml. This solution can then be diluted into other aqueous buffers and stored at -20°C for future use.
                            Storage & Stability
                                The lyophilized powder, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted rHu IL-10 should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long term storage, it is recommended to add a carrier protein (0.1% HSA or BSA). Please avoid repeated freeze / thaw cycles.
