Description
BMP-4 regulates similarly to its nearest homologue BMP-2, diverse fundamental processes during embryonic development. BMP-4 and other BMP proteins are members of the transforming growth factor-beta (TGF-beta) superfamily of proteins and have great potential for medical therapeutic applications, in particular because they allow or at least accelerate the ossification of extensive bone lesions. Bone morphogenetic proteins were originally identified by an ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extraskeletal site. This particular family member plays an important role in the onset of endochondral bone formation in humans, and a reduction in expression has been associated with a variety of bone diseases, including the heritable disorder Fibrodysplasia Ossificans Progressiva. Alternative splicing in the 5 untranslated region of this gene has been described and three variants are described, all encoding an identical protein. Human recombinant BMP-2 produced in HEK cells is a glycosylated disulfide linked homodimer with a total molecular weight of 34 kDa. It has been purified by proprietary chromatographic techniques.

                            Activity
                                rHu BMP-4 is fully biologically active when compared to a standard. The specific activity was determined by the dose-dependent induction of alkaline phosphatase production in the ATDC-5 cell line (Mouse chondrogenic cell line) and is typically 2-10 ng/ml.

                                Less than 1 EU/μg.
                            Formulation
                                Sterile-filtered, white, lyophilized (freeze-dried) powder. Lyophilized from a concentrated protein solution (1 mg/ml) in 1xPBS.
                            Reconstitution
                                Please centrifuge product before opening the vial. The lyophilized protein should be reconstituted in sterile, ultra-pure water to a concentration of 0.1-1.0 mg/ml. This solution can then be diluted into water or other aqueous buffers and stored at -20°C for future use.
                            Storage & Stability
                                The lyophilized protein, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted rHu BMP-4 should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long-term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid repeated freeze-thaw cycles.
