Human Bone Morphogenetic Protein-2 (BMP-2) is a member of the TGF-beta superfamily of cytokines that effect bone and cartilage formation. Human BMP-2 is a disulphide linked homodimeric protein with an apparent molecular weight of 26 kDa. BMP-2 regulates similarly to its nearest homologue BMP-4, diverse fundamental processes during embryonic development, including induction of bone and cartilage growth and regeneration, cardiac cell differentiation and epithelial to mesenchymal transition. BMP-2 promotes osteogenic and chondrogenic differentiation from mesenchymal cells, stem cells, and vascular smooth muscle cells. BMP-2 and other BMP proteins have great potential for medical therapeutic applications, in particular because they allow or at least accelerate the ossification of extensive bone lesions.

                            Activity
                                Each is tested for bioactivity and verified to have comparable activity to a commercial source. The bioactivity of recombinant human BMP-2 was determined by its dose-dependent induction of alkaline phosphatase production by ATDC-5 mouse chondrogenic cells. The ED50 value for this batch of BMP-2 was determined to be below 75 ng/ml, corresponding to a specific activity of >13,300 units/mg. Optimal concentration should be determined for specific applications and cell lines.

                            Formulation
                                Lyophilized from a concentrated, sterile protein solution containing 25 mM MES (pH 6.0).
                            Reconstitution
                                Please centrifuge product briefly before opening vial. The lyophilized protein should be reconstituted in sterile, ultrapure water to a concentration no less than 50 μg/ml. This solution can then be diluted into other aqueous buffers and stored at -20°C for future use. Due to the protein nature, dimmers and multimers may be observed. Optimal concentration should be determined for specific application and cell lines.
                            Storage & Stability
                                The lyophilized protein, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted rHu BMP-2 should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long-term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid repeated freeze-thaw cycles.
