Description
IL-4 is a pleiotropic cytokine produced by activated T cells. IL-4 is a potent lymphoid cell growth factor, which stimulates the growth and survivability of certain B-cells and T-Cells. It is a ligand for the Interleukin-4 receptor. The IL-4 receptor also binds to IL-13, which may contribute to many overlapping functions of this cytokine and IL-13. STAT6, a signal transducer and activator of transcription, has been shown to play a central role in mediating the immune regulatory signal of this cytokine. This gene, IL-3, IL-5, IL-13, and CSF-2 form a cytokine gene cluster on chromosome 5q, with this gene particularly close to IL-13. IL-4, IL-13 and IL-5 are found to be regulated coordinately by several long-range regulatory elements in an over 120 kilobase range on the chromosome. Two alternatively spliced transcript variants of this gene encoding distinct isoforms have been reported. Human recombinant Interleukin-4 produced in HEK cells is a glycosylated monomer, having a molecular weight range of 14-19 kDa due to glycosylation. It has been purified by proprietary chromatographic techniques.

                            Activity
rHu IL-4 is fully biologically active when compared to standards. The activity was determined by the dose-dependent stimulation of the proliferation of human TF-1 cells (human erythroleukemic indicator cell line) and is typically 0.1-0.5 ng/ml.
                            Endotoxin Level
                                Less than 1 EU/μg.
                            Formulation
                                Sterile-filtered, white, lyophilized (freeze-dried)powder. Lyophilized from a concentrated protein solution (1 mg/ml) in 1xPBS.
                            Reconstitution
Please centrifuge product briefly before opening vial. The lyophilized protein should be reconstituted with sterile, ultra-pure water to a concentration of 0.1-1.0 mg/ml. This solution can then be diluted into other aqueous buffers and stored at -20°C for future use.
                            Storage & Stability
The lyophilized powder, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted rHu IL-4 should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long term storage, it is recommended to add a carrier protein (0,1% HSA or BSA). Please avoid repeated freeze / thaw cycles.
