Primary cells are spread in 6-well plates. Infect primary cells with GFP lentivirus, specify cell MOI, and after specifying the minimum lethal concentration for cellular drug screening, infect cells with immortalized lentivirus. On the second day of infection, fresh cell culture medium is replaced and culture continued. After 72 hours of infection, fresh cell culture medium with screening resistance is replaced and culture is continued. After obtaining screen-resistant positive cell clones, the immortalization test can be performed.
