Description
The TOP2A gene, located at 17q21-q22 encodes topoisomerase II-α, a key enzyme in DNA replication, cell cycle progression, and chromosome segregation. As a key enzyme in DNA replication, TOP2A protein is the molecular target for many inhibitors. The TOP2A gene is located telomeric to the HER-2 oncogene, which is located in the 17q11.2-q12 region. HER-2 is one member of a family of transmembrane protein receptors. The close proximity of HER-2, TOP2A, and other genes in the 17q region, suggest a potential relationship between these genes. The TOP2A gene has also been shown to be co-amplified with HER-2 in cell lines and in human breast cancers.
