Cyclin D1 (CCND1) is a critical regulator of the cell cycle, facilitating the transition from the G1 phase to the S phase in collaboration with its binding partners, CDK4 and CDK6. This process occurs through the phosphorylation and subsequent inactivation of the retinoblastoma protein (pRb), a key tumor suppressor. First identified in 1991, CCND1 has gained importance in cancer research due to its amplification in approximately 9-30% of breast cancer cases.
