DSS Induced Colitis Model

Dextran sulfate sodium (DSS) is a polymer of sulfated polysaccharide with a highly variable molecular weight, ranging from 5 to 1400 kDa. DSS induced colitis mouse and rat are the most widely used models due to their simplicity of establishing the model as well as their reproducibility of phenotypes. Animals given 5% DSS drinking water for 5 days exihibit typical symptoms including body weight loss, bloody diarrhea, shortening of colon length, ulceration and intestinal inflammation. And the pathogenesis of DSS induced colitis may be involved in direct hyperosmotic damage to epithelial cells. In addition, chronic and relapsing colitis models can be achieved by modifying the concentration of DSS and the frequency of DSS administration. Study examples

Grim19 prevented induction of DSS induced colitis. (A) Mock or GRIM19 overexpression vectors were administered to HEK293 cell and GRIM19 transgenic mice overexpresssing Grim19 were generated on a C57BL/6 background. Lystes of Mock vector or GRIM19 vector transfected HEK293 cells were analyzed for GRIM19 protein expression using western blotting with and anti-GRIM19 antibody. Transgene expression were detected by PCR products. (B) Change in body weight during the disease process. The DAI score in mice with DSS induced colitis decreased significantly.
