DSS Induced Colitis Model

Inquiry

Dextran sulfate sodium (DSS) is a polymer of sulfated polysaccharide with a highly variable molecular weight, ranging from 5 to 1400 kDa. DSS induced colitis mouse and rat are the most widely used models due to their simplicity of establishing the model as well as their reproducibility of phenotypes. Animals given 5% DSS drinking water for 5 days exihibit typical symptoms including body weight loss, bloody diarrhea, shortening of colon length, ulceration and intestinal inflammation. And the pathogenesis of DSS induced colitis may be involved in direct hyperosmotic damage to epithelial cells. In addition, chronic and relapsing colitis models can be achieved by modifying the concentration of DSS and the frequency of DSS administration. Creative Bioarray specializes in providing customized pharmacodynamic research services to help customers assess the efficacy of drug candidates and study the associated pathological mechanisms of IBD through this model.

Our Capabilities

  • We utilize DSS induced colitis model to test the efficacy of drug candidates targeting IBD.
  • We assess the severity of IBD through clinical analysis including weight loss, diarrhea and occult blood in stool.
  • We evaluate various cytokines through IHC, ELISA, etc.

Assays available

  • Clinical analysis
  • Histopathological evaluation
  • Analysis of mRNA and protein levels
  • Cytokine/chemokine analysis
  • Immunohistochemistry

With extensive experience in the field of IBD, we are confident to help you to overcome any upcoming challenges. Our experts are fully capable of customizing our protocols and assays to meet your specific needs. With our help, we wish to facilitate your research with high efficiency.

Study examples

Grim19 prevented induction of DSS induced colitis. (A) Mock or GRIM19 overexpression vectors were administered to HEK293 cell and GRIM19 transgenic mice overexpresssing Grim19 were generated on a C57BL/6 background. Lystes of Mock vector or GRIM19 vector transfected HEK293 cells were analyzed for GRIM19 protein expression using western blotting with and anti-GRIM19 antibody. Transgene expression were detected by PCR products. (B) Change in body weight during the disease process. The DAI score in mice with DSS induced colitis decreased significantly. (n = 5) (C) Colon length. (n = 5) (D) Histopathological changes in the colon. Sections were stained with hematoxylin and eosin (H&E). Scale bar = 100um Values are means ± SD of three independent experiments.(n = 5) **P<0.01, ***P<0.001.Figure. 2. Grim19 prevented induction of DSS induced colitis.
