Description
Under normal physiological conditions, cells express hypoxanthine-guanine phosphoribosyltransferase (HGPRT), which catalyzes the conversion of 6-thioguanine (6-TG) into toxic nucleotides such as nucleoside-5'-monophosphates (NMPs). These metabolites are incorporated into DNA, leading to cell death. However, upon exposure to carcinogens and/or mutagens, mutations may occur in the structural gene for HGPRT located on the X chromosome, resulting in a loss of HGPRT activity. These mutant cells become resistant to 6-TG and are able to survive and proliferate in selective medium containing 6-TG.
In this assay, cultured mammalian cells are exposed to the test substance—with or without an exogenous metabolic activation system—for a defined period. Following treatment, cells are subcultured and then plated in selective medium containing 6-TG. Mutant cells capable of 6-TG resistance will form discrete colonies, which are counted to determine the mutation frequency, thereby evaluating the mutagenic potential of the test article.
