Fluorescent labeling dyes are commonly used in protein binding or localization studies. These dyes can be incorporated at several positions of peptide chains. Frequently, they are incorporated at the N-terminus or at Cys/Lys side chains. Fluorescent dyes allow scientists to detect a specific stimulus or particular components in complex bio-assemblies with high sensitivity and selectivity.
Fluorescent dyes are usually used to modify amino groups, sulfhydryl groups, carboxyl groups and other biological molecules. Among these reactive dyes, amine-reactive fluorescent dyes are most often used to prepare bioconjugates for immunochemistry, fluorescence in situ hybridization (FISH), cell tracing, receptor labeling and fluorescent analog cytochemistry. Thiol-reactive fluorescent dyes are usually used to probe biological structures, functions and monitor the changes in protein conformation. Carbonyl-reactive fluorescent dyes are widely used for modifications of carbohydrates, glycoproteins and nucleic acids and ketones into the water-soluble biopolymers.
