It is sufficient to inoculate the cells at the density shown in the basic data of the cell line or at the ratio of the diluted plates. Too few cells or too much dilution is also an important reason for the failure of cell growth. According to our experience: for cells with a doubling time of 24 h, a passaging ratio of 1:6-1:12 is appropriate; for cells with a doubling time of 24-48 h, a passaging ratio of 1:3-1:8 is appropriate; for cells with a doubling time of more than 48 h, a passaging ratio of 1:2-1:4 is appropriate.
