Introduction to Fluorescent In Situ Hybridization                                                                         [ TOP ]

Fluorescence in situ hybridization (FISH) techniques allow specific nucleic acid sequences to be detected in morphologically preserved chromosomes, cells or tissue sections. In combination with immunocytochemistry, Fluorescence in situ hybridization can relate microscopic topological information to gene activity at the DNA, mRNA, and protein level. FISH has become a molecular diagnostic technique utilizing fluorescently labeled DNA probes to detect or confirm gene or chromosome abnormalities. The sample DNA (metaphase chromosomes or interphase nuclei) is first denatured, a fluorescently labeled probe of interest is then added to the denatured sample mixture and hybridizes with the sample DNA at the target site as it re-anneals back into a double helix. The probe signal can then be seen through a fluorescent microscope and the sample DNA can be scored for the presence or absence of the signal. Unlike most other techniques used to study chromosomes, FISH does not have to be performed on cells that are actively dividing. This makes it a very versatile procedure. This technique is useful for identifying chromosomal abnormalities, gene mapping, characterizing somatic cell hybrids, identifying amplified genes and studying the mechanism of rearrangements. RNA ISH is used to measure and localize mRNAs and other transcripts within tissue sections or whole mounts.


 

The procedure to perform FISH test                                                                                           [ TOP ]

FISH is useful, for example, to help a researcher identify where a particular gene falls within an individual's chromosomes. Here's how it works:

o  Make a probe complementary to the known sequence. When making the probe, label it with a fluorescent marker, e.g. fluorescein, by incorporating nucleotides that have the marker attached to them.

o  Put the chromosomes on a microscope slide and denature them.

o  Denature the probe and add it to the microscope slide, allowing the probe hybridize to its complementary site.

o  Wash off the excess probe and observe the chromosomes under a fluorescent microscope. The probe will show as one or more fluorescent signals in the microscope, depending on how many sites it can hybridize with.

FISH utilize fluorescent probes that bind to only specific sequence of certain chromosome. Fluorescence microscopy can be used to find out where the fluorescent probe bound to the chromosomes. So FISH is developed for genetic counselling, medicine, and species identification. The applications of FISH for diagnostics and scientific research are as follows:

RNA FISH is a method of detecting and quantifying mRNA or microRNA molecules in a thin layer of tissue or cell sample. Targets can be reliably imaged through the application of multiple short singly labeled oligonucleotide probes. In this context, it can help define the spatial-temporal patterns of gene expression within cells and tissues.

Customer provided tissues. Here we strongly suggest using Carnoy's fixative [Ethanol, chloroform, and acetic acid (6:3:1)] to fix the tissues, and ship the tissues in this fixative. Our experience indicates that this is the best fixative that preserves mRNA the best and allows the most beautiful RNA ISH results. Also, tissues can be shipped in this buffer at room temperature. Please be advised that formalin as well as Qiagen RNA stabilizer is NOT a good choice.

Result Interpretation in a final report

Comprehensive final report, in which our pathologists will score mRNA expression level for each tissue and provide 3-5 representative images for each tissue sample. This format is necessary for studies that require detailed observations and interpretation. We will use FDA scoring staining system. Strong staining was given a score of 3 (+), moderate staining a score of 2 (+), and weak staining a score of 1 (+). Tissues lacking staining entirely were scored 0 (-).

Brief final report, in which our pathologists score the signal present in major cell types of each tissue sample and present the results in a table. Customers can choose to include selective images for each sample with this reporting format.
