Description
IL-17 is a potent proinflammatory cytokine produced by activated memory T cells. There are at least six members of the IL-17 family in humans and in mice. As IL-17 shares properties with IL-1 and TNF-alpha. It may induce joint inflammation and bone and cartilage destruction and high levels of IL-17 are associated with several chronic inflammatory diseases including rheumatoid arthritis, psoriasis and multiple sclerosis. Blocking of IL-17 with specific inhibitors provides a protective inhibition of cartilage and bone degradation. It increases IL-6 production, induces collagen degradation and decreases collagen synthesis by synovium and cartilage and proteoglycan synthesis in cartilage. IL-17 is also able to increase bone destruction and reduce its formation. It regulates the activities of NF-kappaB and mitogen-activated protein kinases and can stimulate the expression cyclooxygenase-2 (PTGS2/COX-2) as well as enhance the production of nitric oxide (NO). Recombinant rat IL-17 produced in E. coli is a homodimeric, non-glycosylated polypeptide chain of 266 amino acids and a molecular mass of 29,956 Dalton. It has been purified by proprietary chromatographic techniques.

Formulation
Lyophilized (freeze-dried), white powder. Lyophilized from a concentrated (1 mg/ml) proten solution in 10 mM Na-citrate (pH 3.0).
Reconstitution
Please centrifuge product briefly before opening vial. The lyophilized protein should be reconstituted in sterile, ultra-pure water to a concentration of 0,1 - 1,0 mg/ml. This solution can then be diluted into other aqueous buffers and stored at -20°C for future use.
Storage & Stability
The lyophilized protein, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted rRa IL-17 should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long-term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid repeated freeze-thaw cycles.
