TIE-1 and TIE-2/Tek comprise a receptor tyrosine kinase (RTK) subfamily with unique structural characteristics: two immunoglobulin-like domains flanking three epidermal growth factor (EGF)- like domains and followed by three fibronectin type III-like repeats in the extracellular region and a split tyrosine kinase domain in the cytoplasmic region. These receptors are expressed primarily on endothelial and hematopoietic progenitor cells and play critical roles in angiogenesis, vasculogenesis and hematopoiesis. Human TIE-2 cDNA encodes a 1124 amino acid (aa) precursor protein with an 18 aa putative signal peptide, a 727 aa extracellular domain and a 354 aa cytoplasmic domain. Two ligands, angiopoietin-1 (Ang-1) and angiopoietin-2 (Ang-2), which bind TIE-2 with high-affinity have been identified. Ang-2 has been reported to act as an antagonist for Ang-1. Recombinant human soluble TIE-2/TEK was fused with a 6x His-tag at the C-terminus. The soluble receptor protein consists of the full extracellular domain (Thr19-Lys745). It contains 731 amino acids and has a molecular weight of 95 kDa.

Formulation
Lyophilized from a steril-filtered protein solution in PBS without any stabilizers.
Reconstitution
Please centrifuge product briefly before opening vial. The lyophilized protein should be reconstituted in ultra-pure, sterile water or aqueous buffers to a concentration of 0.1 - 1.0 mg/ml. This solution can then be diluted into other aqueous buffers and stored at -20°C for future use.
Storage & Stability
The lyophilized protein, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted rHu sTIE-2 should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long-term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid repeated freeze-thaw cycles.
