Description
Tumor necrosis factors are cytokines involved in systemic inflammation and are member of a group of cytokines that all stimulate the acute phase reaction. TNF is mainly secreted by macrophages and is a potent lymphoid factor which exerts cytotoxic effects on a wide range of tumor cells and certain other target cells. TNF causes apoptotic cell death, cellular proliferation, differentiation, inflammation, tumorigenesis and viral replication, TNF is also involved in lipid metabolism, and coagulation. TNFs primary role is in the regulation of immune cells. Dysregulation and, in particular, overproduction of TNF have been implicated in a variety of human diseases such as autoimmune diseases, insulin resistance, and cancer. Recombinant human Tumor Necrosis Factor-alpha (rHu TNF-alpha) produced in HEK cells is a glycosylated non-disulfide linked homotrimer, having a total molecular weight of 17 kDa. It has been purified by proprietary chromatographic techniques.

Activity
rHu TNF-alpha is fully biologically active when compared to standards. The specific activity was determined by the dose-dependent cytotoxity of the TNF alpha sensitive cell line L-929 in the presence of Actinomycin D and is typically 0.05-0.5 ng/ml.

Less than 1 EU/μg.
Formulation
Sterile-filtered, white, lyophilized (freeze-dried) powder. Lyophilized from a concentrated protein solution (1 mg/ml) in 1xPBS.
Reconstitution
Please centrifuge product briefly before opening the vial. The lyophilized rHu TNF- alpha should be reconstituted in sterile, ultra-pure water to a concentration of 0.1-1.0 mg/ml. This solution can then be further diluted into other aqueous buffers and stored at -20°C for future use.
Storage & Stability
The lyophilized rHu TNF-alpha, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted rHu TNF-alpha should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long-term storage, we recommend the addition of a carrier protein (0.1% HSA or BSA). Avoid repeated freeze / thaw cycles.
