Description
Recombinant Human Thrombopoietin (rHu TPO, Mpl-ligand, MGDF) is a glycoprotein hormone produced mainly by the liver and the kidney. It stimulates the proliferation and maturation of megakaryocytes and promotes increased circulating levels of platelets produced by the bone marrow in vivo. |Human recombinant Thrombopoietin produced in CHO cells consists of 332 amino acid residues with a predicted molecular mass of approximately 35 kDa. As a result of glycosylation, the recombinant protein migrates with an apparent molecular mass of 80±10 kDa in SDS-PAGE.

Activity
rHuTPO is fully biologically active when compared to a standard. The ED50, as determined by the dose-dependant stimulation of MO7e cells, is ~1 ng/ml, corresponding to a specific activity of ~1 x 10^6 IU/mg.

Formulation
Lyophilized from a sterile-filtered protein solution containing 20 mM sodium citrate (pH 6.9).
Reconstitution
Please centrifuge product briefly before opening vial. The lyophilized protein should be reconstituted in sterile, ultra-pure water to a concentration of 0,1 - 1,0 mg/ml. This solution can then be diluted into other aqueous buffers and stored at -20°C for future use.
Storage & Stability
The lyophilized protein, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted rHu TPO should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long-term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Avoid repeated freeze-thaw cycles.
