Description
RANTES (Regulated upon Activation, Normal T cell Expressed and presumably Secreted) is a member of the b (CC) family of chemokines. It is produced by synovial fibroblasts, monocytes, T lymphocytes, eosinophils, mast cells, platelets, epithelial cells, and some tumor cells. RANTES is a chemoattractant for monocytes, T cells, basophils, eosinophils, NK cells, dendritic cells and mast cells. In addition, RANTES induces activation of basophils, eosinophils and NK cells and it shows HIV-inhibitory effects. RANTES binds to CCR1, CCR3, CCR5, DARC and US28 receptors. Human RANTES cDNA encodes a highly basic 91 amino acid residue precursor with a 23 amino acid residue hydrophobic signal peptide that is cleaved to generate the 68 amino acid residue mature protein. The recombinant mature protein has a predictedmolecular mass of 7.8 kDa.

Activity
Activity was determined by monocyte chemotaxis and ability to chemoattract mouse BaF/3 cells transfected with the human CCR5 receptor. The ED50 for these effects were typically 10 - 30 ng/mL and 1- 5 ng/mL, respectively.
Endotoxin Level
Endotoxin level less than 0.1 ng per 1 mg of the cytokine as determined by the LAL method.
Formulation
Lyophilized from a sterile solution in 30% acetonitrile, 0.1% TFA containing 50 mg of bovine serum albumin per 1 mg of cytokine.
Reconstitution
It is recommended that sterile PBS containing at least 0.1% human serum albumin or bovine serum albumin be added to the vial to prepare a stock solution of no less than 10 mg/mL of the cytokine.
Storage & Stability
Lyophilized samples are stable for greater than six months at -20°C to -70°C. Reconstituted human CCL5 can be stored under sterile conditions at 2°C to 4°C for one month or at -20°C to -70°C for three months without detectable loss of activity. Avoid repeated freeze-thaw cycles.
